Your question isn't clear, and you haven't supplied enough information.

Do you know what the variable profile is (i.e. how it changes with time) before running the simulation?
If so, just define it in the MATLAB workspace and import it through an Inport or a From Workspace block.

If you want to change a value asynchronously then you have already shown the basic command:
set_param('modelname/variable','value',' ');
Although you need to look at the doc for set_param to see how to use it correctly.
