str = "hELLO THERE!";

For a string of this form

StringReplacePart[
ToLowerCase[str],
ToUpperCase[StringTake[str, 1]],
1]

"Hello there!"

For this specific string

StringReplace[
ToLowerCase[str],
"h" -> "H", 1]

"Hello there!"
